First Europe Trip -- advice where to start please

I'm new to the site and this will be my first Europe trip. I'm 30 and going solo. I've narrowed my choices to Amsterdam to Barcelona and the Green Island Hopping. I'm leaning towards the first as i can spend time with a relative in Madrid for a bit hopefully but the latter sounds so appealing; however, as much as I would like to hang loose and meet lots of people, I'd like to also experience some culture.
Any opinions from people who've been to any of these tours?
